Abstract

The aim of this study was to compare the isolated metagenomic DNA of mangrove soil in Suwung Kauh Beach, Bali by utilizing suspended bentonite of NaCl, MgCl2 and CaCl2. The metagenomic DNA isolation was performed by directly lysis method from soil with lysis buffer and heat-shock, and then continued by DNA extraction with suspended bentonite of NaCl, MgCl2 and CaCl2. Preparation of suspended bentonite was done in 1 M NaCl, MgCl2 and CaCl2. The results of metagenomic DNA isolation were analysed by gel agarose electrophoresis and spectrophotometer Nano at wave length 230, 260, and 280 nm. The results of gel agarose electrophoresis showed that DNA was not detected in the first elution from all suspended bentonite, but DNA was only detected in the first supernatant before elution from Na-bentonite and Mg-bentonite suspended. The DNA bands were most thick in the supernatant before the elution of Na-bentonite. This result indicated that Na-bentonite adsorption to metagenomic DNA was lower than the other bentonite suspended. The DNA band was not detected in the first supernatant and the first elution from Ca-bentonite that indicated that DNA adsorption was stronger than the other and was not breakable from this suspended. The results of spectrophotometer nano showed that the metagenomic DNA isolated by suspended bentonite of NaCl, MgCl2 and CaCl2 were contaminated by humic acid and protein.

Abstract

A research on heavy metal speciation and bioavailability of Cu and Zn in soil of organic agriculture in Bedugul area has been carried out. This study aimed to determine the total metal content, speciation, and bioavailability of Cu and Zn in the soil of organic agriculture aged 5 and 4 years in Bedugul area. The total metal content was performed by the used of a digestion method with a mixture of HNO3 and HCl (3:1) in the ultrasonic bath and followed by heating on the hotplate. Speciation and bioavailability of Cu and Zn was conducted using SET (Sequential Extraction Technique). The content of Cu and Zn in the total sample was analyzed using the Atomic Absorption Spectrophotometer (AAS) with a calibration curve method. The total metal concentrations of Cu in the organic agricultural land of aged 5 and 4 years and were 41.8808-49.1060 mg/kg and 22.8544-41.6616 mg/kg  whereas the concentrations of Zn  in the organic agricultural land of aged 5 and 4 years were 62.9273-73.3729 mg/kg and 58.0970-73.9987 mg/kg respectively. The speciation of Cu obtained in in the organic agricultural land of aged 5 and 4 years was ranged from 6-17% and 3-14% in the EFLE fraction, from 9 to 37% and 6 to 24%  in the Fe/Mn oxides fraction, and ranged from 27 to 53% and 41 to 69% in the resistant fraction (non bioavailable). The Zn speciation found in the organic agricultural land of aged 5 and 4 years was ranged from 6 to 9% and 7 to 10% the EFLE fraction (bioavailable), from 15 to 22% and 11 to 25% in the Fe/Mn oxides and organic (potentially bioavailable) fractions and ranged from 46 to58% and 40 to 53%  in the silicate fraction (non bioavailable).

Abstract

This research aimed to determine the effectivity of soursop leaf extract to repair beta pancreatic cells damaged in Wistar rats induced by alloxan. This experiment used randomized posttest-only control group design. In this experiment, 24 Wistar rats were used which divided into four groups, control group (K), and three treatment groups (P). P1 was the group tretment with a dose of 50 mg/kg bw/day, P2 with a dose of 100 mg/kg bw/day,  and P3 was the group treatment with a dose of 150 mg/kg bw/day extract. All groups were induced by alloxan with a dose of 150 mg/kg bw to obtain condition of hyperglycemia. The results showed that blood glucose levels decreased after treatment with the extracts. Glucose levels in group K, P1, P2, and P3 were 208.0 mg/dL; 177.5mg/dL; 164.7 mg/L and 137.0 mg/dL respectively. After 2 weeks of treatment, the level of 8-OHdG and AGEs were also reduced. The lowest level was observed in P3 treatment which the level of 8-OHdG 1.664ng.mL and 0.033 mol/L for AGEs level. In conclusion the soursop leaf (Annona muricata) extract given at a dose of 150 mg/kg bw/day can improve beta pancreatic cells resulted from antioxidant activity content in the extract which function as antihyperglycemia. Based on GC-MS spectra analysis,  the the soursop leaf (Annona muricata) extract consisted of caryophyllene, ethyl tetradekanoat, acid 2-cyclohexane-1-one, 4-hydroxy-3,5,6-trimethyl-4-(3-oxo-1-butenyl), hexa ethyl decanoate , phytol, thiazole-2,4 (3H, 5H) _dion, 5 benzilideno-3 [etifenilamino) metal], 9-octadecanoic acid, ethyl linoteat, ethyl oleate and ethyl octadecanoic.

Abstract

Activated clay can be used as an adsorbent to adsorb phosphate ions in solution. The aims of this research are to study the characteristics of the clay before and after activated (covering the surface acidity, surface area and basal spacing) and to study the adsorption capacity of activated clay to phosphate solution. The results showed, the activated clay gave surface acidity of 0.7202 mmol/g and 2.0018 mmol/g, surface area of 12.6602 m2/g dan 25.6101 m2/g, and d-spacing of montmorillonite was d (Å) = 0.12. The optimum condition for phosphate adsorption by this clay was found at 15 minutes contact time, pH 3, and  concentration of 70 ppm yielding phosphate adsorption capacity of 3.0349 mg/g following the Langmuir isotherm.

Abstract

The research was conducted to observe diphenylcarbazide immobilization on silica gel from rice husk ash (Si-CPTMS-DPZida) with sol-gel technique. The aim of this research was to modify silica gel from rice husk ash by immobilization with diphenylcarbazide using sol-gel technique. This research included the modification and characterization of silica gel. The characterizations of silica gel were done by using infrared spectrophotometer (FTIR) and X-ray diffraction (X-RD). The results showed that the immobilized diphenylcarbazide silica has been formmed which is indicated by the appearance of Si-OH functional group in the peak at wave number 914,26 cm-1, Si-O-Si at 1091,71 cm-1, the stretching vibration of NH group at 3645 cm-1 and proved in 1492 cm-1 which is the bending vibrations of the NH group, CH (aromatic) at  802.39 cm-1, vibrational bending C = C (aromatic) at 1612 cm-1, vibration CN group at 1310 cm-1, and the vibrations of the C = O group in the area of 1685 cm-1. The X-RD data showed that the structure of Si-CPTMS-DPZida is amorphous.

Abstract

The peel of banana kepok fruit (Musa paradisiaca Linn.) contains compound that function as an antioxidant in biodiesel. The purposes of this study were to determine the peroxide number, acid number, and FFA content of biodiesel from waste palm oil with addition of chloroform extract (EKKBPK). Biodiesel was synthesized using esterification reaction of wasted palm oil with methanol (10:1) and by transesterification reaction (4:1), the extract of peel of banana kepok fruit was theen added with various concentration of 0,1 % w/v (A1); 0,2 % w/v (A2); and 0,3 % w/v (A3) to the reaction mixtures. The peroxide number, acid number, and FFA content of synthesized biodiesel obtained in this research were 9.3591 meq peroxide/1000 g; 0.7221 mg KOH/g; and 0.33% repectively; biodiesel without addition of EKKBPK (12.2746 meq peroxide/1000 g; 0.6492 mg KOH/g; and 0.30%); and biodiesel with addition of EKKBPK 0.1 % w/v (11,1878 meq peroxide/1000 g; 0,4814 mg KOH/g; and 0.22%); 0.2 % w/v (9.7508 meq peroxide/1000 g; 0.3501 mg KOH/g; and 0.16%); and 0.3 % w/v (12.2512 meq peroxide/1000 g; 0.5217 mg KOH/g; and 0.24%). The acid numbers of those biodiesel were in acordance to the acid number of SNI 04-7182-2006. The addition of 0.2 % w/v EKKBPK on biodiesel showed the lowest peroxide number, acid number, and FFA content threfore it was the most effective in inhibiting the oxidation reaction of biodiesel.

Abstract

DNA content in total honey microorganism can be used to study DNA fingerprint of honey. Metagenomic is a new approach to analyse complex genomes from unculturable total microorganism. The aim of this research was to get DNA metagenomic isolated directly from honey samples enriched with microorganisms growing media LB (Luria-Bertani) and without enrichment of microorganisms growing media LB in order to study DNA fingerprint of honey for the next research. The study was started with incubation of the LB enriched honey samples and honey without LB samples at variation time of 0, 24, 48, and 72 hours. Following the incubation process, the DNA metagenomic form both LB enriched and unenriched honey samples were isolated with direct cell lysis method. The DNA obtained was analyzed by spectrophotometer UV-Vis and agarose gel electrophoresis.  The result showed that DNA metagenomic can be isolated from LB enriched honey and without LB enriched honey samples. Analysis using the agarose gel electrophoresis indicated that the metagenomic DNA obtained from both honey samples was still contaminated with RNA. The purity ratio of metagenomic DNA from both honey samples analysed using UV-Vis Spectrophotometer at ? 260/280 nm was between 1.2 to 1.6. This ratio suggested that the metagenomic DNA isolated from both samples were contaminated with protein and fenol.

Abstract

A research on synthesis diphenylcarbazone immobilized silica gel from rice husk ash by sol-gel technique has been conducted. The purpose of this research was to synthesize the diphenylcarbazone immobilized silica gel from rice husk ash by sol-gel technique.  This research included modification of silica gel of rice husk ash and silica gel characterization by IR spectra and X-RD. The results showed that the diphenylcarbazone immobilized silica gel has been successfully synthesized. It can be seen from the FTIR spectrum that silica-imobilized revealed peaks at wave numbers of ±1087.85cm-1 indicating the presence of Si-O-Si. The peak seen at wave numbers of  ±952.84cm-1 indicating the presence of Si-O, at ±3361.93cm-1due to the Si-OH bond which was the main functional group  of silica gel. Another peaks at ±3313.71cm-1, ±800.46cm-1, ±1602.85cm-1,±1700 cm-1, and ±1602.85cm-1 suggesting the presence of N-H, aromatic C-H (substituted benzene), aromatic C=C, N=N and C=O stretching. X-RD data showed that the structure of diphenilcarbazone immobilized silica gel was amorphous.

Abstract

These researchs aim to know of contain total flavonoid, total phenol and antioxidant activity in vitro of gayam stem bark of each extract (Inocarpus fagiferus Fosb). The result of extraction 1 kg stem bark of gayam with 8500 mL ethanol (5 x 1700 mL) to yield 30 g of concentrated extract its brown red. The partition of 15 g concentrated ethanol extract with chloroform, n-buthanol, and water to yields of 1.5g yellowish concentrated chloroform extract,  5.87 g brown red concentrated n-buthanol extract and 5.63 g concentrated water extract respectively. The result of phytochemical test of fourth extract shown that the ethanol, n-buthanol, and water extracts qualitatively contain of flavonoid and phenol compounds, while the chloroform extract only to contain phenol compound with much total flavonoid in ethanol extract 0.14 % and less at n-buthanol extract is 0.09 %. The result of antioxidant activity with DPPH method shown that water and  n-buthanol extract of gayam stem bark have value IC50 was 20 ppm smaller than  IC50 of vitamin E was 25 ppm its mean the water and  n-buthanol extract of gayam stem bark have the highest potent scavenger of free radical. The result of antioxidant activity with lipid peroxide-ammonium thiocyanate method shown the extract n-buthanol extract have the highest potent  inhibition of formed lipid peroxide (63.04%), it is higher than antioxidant activity of vitamin E (50.39%).

Abstract

7-hydroxy-6-methoxy coumarin (scopoletin) has been isolated from the bark of Chisocheton celebicus (Meliacee) using several chromatographic techniques.  The chemical structure of 7-hydroxy-6-methoxy coumarin was identified on the basis of spectroscopic data including UV, IR, 1D-NMR, 2D-NMR and mass along with by comparison with those spectral data previously reported. The discovery of scopoletin from the bark C. celebicus reported for the first time in this study.
